- Attualmente, l’uso dei rifiuti agricoli nella produzione animale è di interesse in tutto il mondo. Pertanto, in questo documento è stato condotto uno studio di otto settimane per studiare gli effetti di un supplemento di Navel Orange Peel Extract (NPE) sulla crescita, la conversione del mangime, la digeribilità dei nutrienti, gli antiossidanti e l’ematologia nei conigli sottoposti a stress da calore. Un totale di 75 conigli svezzati sono stati assegnati in modo casuale a cinque gruppi. Il primo gruppo era allevato in inverno (tempo mite) e alimentato con mangime in pellet non trattato (W-NPE-0; controllo).
- Gli altri quattro gruppi sono stati allevati in estate (clima caldo) e alimentati con una dieta di controllo integrata con 0 (S-NPE-0), 250 (S-NPE-250), 500 (S-NPE-500) o 1000 (S- NPE-1000) mg NPE / kg dieta. I risultati indicano che i conigli sottoposti a stress termico (S-NPE-0) avevano un consumo di mangime, efficienza di crescita, indici ematologici, profilo lipidico sierico e stato antiossidante significativamente più bassi, ma una maggiore perossidazione lipidica rispetto al gruppo W-NPE-0.
- Tuttavia, il peso finale e l’assunzione di mangime più elevati sono stati registrati nel gruppo S-NPE-1000 rispetto al gruppo S-NPE-0. Inoltre, l’integrazione di NPE nella dieta dei conigli in crescita, in particolare nel gruppo S-NPE-1000, ha migliorato gli indicatori ematologici e antiossidanti. In conclusione, l’integrazione di NPE nella dieta dei conigli in crescita può essere utilizzata per alleviare efficacemente gli effetti deleteri dello stress termico cronico sulle prestazioni, sulle caratteristiche ematobiochimiche e sulla stabilità ossidativa.
Effetto del Clostridium butyricum e del cocktail di batteriofagi sulla crescita, biochimica del siero , attività degli enzimi digestivi, morfologia intestinale, risposta immunitaria e microflora intestinale dei conigli

Lo scopo di questo studio era di valutare l’effetto dell’integrazione alimentare con Clostridium butyricum (CB) e cocktail di batteriofagi (BP) sull’efficienza della crescita, parametri biochimici sierici, enzimi digestivi e ossidasi intestinali, morfologia intestinale, risposta immunitaria e microflora cecale nei conigli. Un totale di 108 conigli neozelandesi (5 settimane di vita) sono stati randomizzati e assegnati equamente a tre gruppi di trattamento dietetico (quattro repliche per trattamento, n = 36/trattamento) : (1) gruppo di controllo (CN) – conigli alimentati con dieta basale; (2) Conigli CB alimentati con una dieta di base integrata con 100 mg/kg di Clostridium butyricum ; e (3) conigli BP alimentati con una dieta basale integrata con 200 mg / kg di frullato dietetico BP, rispettivamente, per 6 settimane.
- Rispetto alla dieta CN, l’inclusione di CB e BP nella dieta ha aumentato il guadagno giornaliero medio (ADG) e l’assunzione media giornaliera di mangime (ADFI) e ha diminuito il rapporto tra mangime e profitto (F / G) dei conigli.
- Inoltre, il CB aumenta l’attività degli enzimi digestivi (α-amilasi e tripsina nell’ileo); anche l’attività della chimotripsina era significativamente aumentata nel duodeno e nel digiuno. La supplementazione di CB ha aumentato significativamente la capacità antiossidante (SOD e GSH-Px) nel digiuno e nell’ileo e ha abbassato il livello di MDA.
- Inoltre, i conigli nutriti con CB presentavano un aumento significativo dell’altezza dei villi (V) e dei rapporti (V / C), ma una ridotta profondità della cripta (C). Inoltre, la supplementazione dietetica con CB ha aumentato significativamente l’espressione ileale delle proteine della giunzione stretta (occludina, ZO-1 e claudina-1) e ha aumentato la produzione di immunoglobulina A secretoria (sIgA).
- Il sequenziamento ad alto rendimento ha mostrato che il microbiota nell’intestino del coniglio era alterato da CB e BP. I diagrammi di Venn e le mappe di calore hanno mostrato che la composizione della comunità microbica intestinale era notevolmente diversa tra conigli alimentati con diete diverse.
- In particolare, CB ha aumentato l’abbondanza relativa di batteri benefici per mantenere l’omeostasi della barriera intestinale, mentre BP ha diminuito l’abbondanza relativa di Gammaproteobacteria, che includeva molti batteri patogeni.
Concentrazioni fisiologicamente rilevanti di hCys mobilitano MeHg dall’albumina sierica di coniglio per formare complessi MeHg-hCys.
- Il metilmercurio (MeHg) è una delle neurotossine più potenti a cui gli esseri umani sono esposti quando mangiano pesce e viene assorbito in modo efficiente attraverso il tratto gastrointestinale nel flusso sanguigno. Tuttavia, le sue interazioni con le proteine plasmatiche, le molecole a basso peso molecolare (SMW) e i globuli rossi non sono completamente comprese, ma sono critiche in quanto determinano se e quanto MeHg raggiunge gli organi bersaglio. Per definire meglio il ruolo che i tioli SMW svolgono nel fornire MeHg a noti trasportatori situati sulla barriera placentare e ematoencefalica, abbiamo utilizzato la cromatografia di esclusione del plasma accoppiata induttivamente con spettroscopia di emissione atomica per analizzare il plasma di coniglio con MeHg aggiunto in assenza e presenza di disciolto Tioli SMW nella fase mobile del buffer PBS.
- Mentre 300 µM di L-metionina non hanno influenzato il legame di MeHg all’albumina sierica di coniglio (RSA), alla cisteina (Cys), all’omocisteina (hCys) e al glutatione (GSH), ha eluito il picco maggiore di Hg in termini di eluizione di SMW . Inoltre, 50 µM di hCys o Cys in fase mobile hanno mobilitato MeHg da RSA nel plasma di coniglio e da RSA pura in soluzione.
- Il picco di Hg eluito nell’intervallo di eluizione dell’SMW (50 µM hCys) è stato identificato mediante spettrometria di massa a ionizzazione elettrospray come complesso MeHg-hCys. Poiché nella barriera ematoencefalica (BBB) sono presenti trasportatori di amminoacidi di tipo L che facilitano l’assorbimento delle specie MeHg-Cys nel cervello, i nostri risultati contribuiscono a stabilire i meccanismi bioinorganici che forniscono MeHg a BBB, che è fondamentale nella previsione degli effetti negativi sulla salute relativi agli organi.
Esposizione di conigli selvatici a Leishmania infantum , Toxoplasma gondii , Anaplasma phagocytophilum e Babesia caballi confermata dal rilevamento di anticorpi nel siero e nell’acqua
- I conigli selvatici ( Oryctolagus cuniculus ) possono essere importanti specie indicatori della presenza di patogeni zoonotici. Pertanto, abbiamo prelevato campioni di sangue da conigli selvatici raccolti dai cacciatori durante la stagione venatoria 2019-2020 sull’isola di Lemno per determinare l’esposizione dei conigli selvatici ai patogeni zoonotici Leishmania infantum , Toxoplasma gondii , Anaplasma phagocytophilum e Babesia caballi , nonché l’umor acqueo per valutarne l’efficacia in termini di sensibilità, specificità, indicatori di affidabilità positivi e negativi. Gli anticorpi contro questi agenti patogeni sono stati rilevati in un test di immunofluorescenza indiretta (IFA). Dei 72 conigli selvatici eIl 4,2%, 5,5%, 18% e 9,7% di quelli inclusi nello studio erano sieropositivi rispettivamente contro L. infantum , T. gondii , A. phagocytophilum e B. caballi .
- Sebbene meno comuni, gli anticorpi sono stati rilevati anche nell’umore acqueo dei conigli selvatici. Il rilevamento di anticorpi nell’umor acqueo ha mostrato una specificità del 100% ma una sensibilità ridotta rispetto al siero, suggerendo che l’umor acqueo può essere utilizzato con successo negli studi epidemiologici per confermare l’esposizione a livello di popolazione, ma ha scarso valore diagnostico a livello individuale.

Round RNA circ_0000644 promuove la progressione del cancro papillare della tiroide attraverso la spugna miR-1205 e la regolazione dell’espressione di E2F3

1. La deregolazione dell’RNA circolare (circRNA) facilita la neoplasia del carcinoma papillare della tiroide (PTC). Lo scopo di questo studio era di determinare la funzione e il meccanismo di circ_0000644 nella regolazione dello sviluppo del PTC. Espressioni di Circ_0000644, microRNA-1205 (miR-1205) ed E2F del fattore di trascrizione 3 (E2F3) sono state rilevate mediante reazione a catena della polimerasi quantitativa in tempo reale (qRT-PCR). Sono stati utilizzati saggi con actinomicina D (ActD) e ribonucleasi R ( RNasi R) per verificare le caratteristiche circolari di circ_0000644. Dopo la soppressione di circ_0000644 , la crescita, la migrazione, l’invasione e l’apoptosi delle cellule PTC sono state valutate utilizzando rispettivamente il kit di conteggio delle cellule -8 (CCK-8), il saggio Transwell e l’analisi citometrica a flusso.
2. La relazione regolatoria tra circ_0000644, E2F3 e miR-1205 è stata confermata nel saggio di immunoprecipitazione dell’RNA (RIP) e nel saggio reporter della doppia luciferasi. Inoltre, gli effetti regolatori di circ_0000644 sul livello della proteina E2F3 sono stati analizzati mediante Western blotting. In PTC, circ_0000644 ha mostrato un’elevata espressione ed era localizzato principalmente nel citoplasma e aveva una struttura stabile. Il contenuto di carbonile proteico è ampiamente utilizzato sia come indicatore dello stress ossidativo che come misura del danno ossidativo. I metodi ampiamente utilizzati di test di carbonilazione proteica utilizzano la reazione di gruppi carbonilici con 2,4-dinitrofenilidrazina (DNPH) per produrre 2,4-dinitrofenilidrazone legato alle proteine. Sia gli oligonucleotidi di DNA sintetizzati in vitro che il DNA cromosomico purificato hanno reagito fortemente con 2,4-DNPH. Il trattamento di estratti cellulari con DNasi + RNasi o streptomicina solfato per precipitare gli acidi nucleici ha ridotto drasticamente il carbonile visibile, mentre l’esposizione alla proteinasi K no.

Il kit commerciale per il rilevamento immunochimico della carbonilazione proteica (OxyBlot di Chemicon / Millipore) raccomanda un’elevata concentrazione di tiolo nel tampone di omogeneizzazione. Abbiamo scoperto che questa raccomandazione porta a un raddoppio artificiale della proteina carbonile, probabilmente a causa di una reazione di Fenton stimolata dal tiolo. Tuttavia, Ca2 + attiva la subunità β-3 del canale del calcio voltaggio-sensibile di tipo L (Ca-β3), che autoregola l’eccitabilità neuronale e svolge un ruolo nello spostamento del GABA dalla neurotrasmissione eccitatoria a quella inibitoria .

Un totale di otto femmine (n = 4 controlli e n = 4 perinatali) e quattro maschi (n = 2 controlli e n = 2 perinatali) sono stati utilizzati come riproduttori per fungere da madri e padri.

Test dell’attività proteasica nell’intestino di una mosca

- - L’intestino è l’organo centrale necessario per digerire il cibo, assorbire i nutrienti e combattere gli aggressori ingeriti con il cibo. L’alterazione della fisiologia intestinale a seguito di danni alla mucosa influisce sulla sua capacità digestiva, che a sua volta influenzerà la crescita, il benessere e persino la sopravvivenza dell’individuo.
  - Pertanto, la valutazione della funzione intestinale comprende, tra l’altro, il monitoraggio della sua integrità, il rinnovamento cellulare, la sua difesa immunitaria, la produzione di ormoni enteroendocrini e la sua capacità digestiva. Qui descriviamo in dettaglio come valutare l’attività delle proteasi secrete nel lume della mosca adulta Drosophila melanogaster . Questo metodo può essere applicato anche agli intestini larvali.   - La principale proteasi di SARS-CoV-2 (M ^pro ) è uno dei bersagli farmacologici più utilizzati per COVID-19. Composti strutturalmente diversi sono stati descritti come inibitori di Mpro, il che solleva la questione della loro specificità target ^.
  - Per chiarire la specificità del bersaglio e il coinvolgimento del bersaglio cellulare degli inibitori dichiarati, abbiamo caratterizzato sistematicamente il loro meccanismo d’azione mediante il test FRET senza cellule, il test di legame dello spostamento termico, il test del lisato cellulare della luciferasi Protease-Glo e il test saggio FlipGFP cellulare.
  - Collettivamente, i nostri risultati hanno mostrato che la maggior parte degli inibitori di M ^pro identificati durante il riutilizzo di farmaci, inclusi ebselen, carmofur, disulfiram e shikonin, sono inibitori della cisteina promiscua che non sono specifici di M ^pro , mentre clorochina, ossitetraciclina, montelukast, candesartan, e il dipiridamolo non ha inibito M ^pro in nessuno dei test testati.   2. Abbiamo utilizzato questo modello per cercare nel database DrugBank e un elenco di due candidati principali (bromocriptina e avoralstat) per il test enzimatico in vitro. Imaging stroboscopico ultraveloce con strisce RF in un microscopio elettronico a trasmissione commerciale

Lo sviluppo della microscopia elettronica ultraveloce (UEM), in particolare l’imaging stroboscopico, ha portato lo studio della dinamica strutturale a un nuovo livello, superando i limiti spaziali della spettroscopia ultraveloce e i limiti di tempo della tradizionale TEM. La combinazione dei concetti che governano entrambe le tecniche ha consentito la visualizzazione diretta della dinamica con risoluzioni spazio-temporali nel regime del picosecondo-nanometro. Qui spostiamo i limiti dell’imaging del fascio di elettroni pulsati tramite la deflessione trasversale indotta da RF basata su un generatore di impulsi a striscia di frequenza regolabile da 200 keV di nuova concezione.

Presentiamo una risoluzione spaziale di 0,2 nm e spieghiamo le mappe di induzione dello spin magnetico mediante microscopia di fase. Presentiamo anche misurazioni della coerenza del fascio ed estendiamo il nostro studio utilizzando le modalità di respirazione del pettine di silicio di eccitazione RF, che forniscono una migliore tempistica tra il treno di impulsi di elettroni e il campo elettrico. È stato inoltre sviluppato un nuovo dispositivo RF con impedenza corrispondente al segnale RF per ridurre al minimo la perdita di potenza di trasmissione ai campioni e le sue prestazioni sono paragonate a quelle di un dispositivo campione convenzionale.
Test del lettore di schede fototermico utilizzando una striscia reattiva in oro colloidale disponibile in commercio per il rilevamento quantitativo rapido dei rischi alimentari
La semplice e veloce striscia reattiva in oro colloidale disponibile in commercio può essere utilizzata solo per il rilevamento qualitativo o semiquantitativo con scarsa rilevabilità e risultati falsi negativi. Questo documento sviluppa una striscia reattiva fototermica che combina la striscia reattiva con un lettore di schede fototermico portatile per ottenere un rilevamento quantitativo con un’eccellente rilevabilità. Secondo l’effetto fototermico prodotto dalle nanoparticelle d’oro (GNP) catturate sulla linea di test, il segnale potrebbe essere catturato da un lettore.

Sono stati studiati tredici rischi alimentari, inclusi residui di farmaci veterinari e residui di pesticidi; la rilevabilità fototermica nei campioni effettivi era di circa 23 (paration metilico), 7 (enrofloxacina), 6 (sarafloxacina), 8 (sulfadiazina), 12 (sulfametazina), 7 (paraquat), 6 (verde malachite), 11 (amantadina), 13 (nitrofurazone), 6 (dietilstilbestrolo), 12 (estriolo), 21 (estrone) e 26 (17β-estradiolo) volte meglio del rilevamento visivo. I nostri risultati hanno mostrato che la striscia reattiva fototermica può essere utilizzata per il rilevamento sensibile, rapido e quantitativo dei residui di rischio alimentare.

Acetato di cellulosa e nanocomposito di cellulosa immobilizzati con il cromoforo DCDHF-idrazone verso una   striscia reattiva intelligente per il rilevamento colorimetrico del dietilclorofosfato come fattore nervoso mimico

L’esposizione agli agenti nervini, che di solito sono gas incolori e inodori, può causare insufficienza d’organo, paralisi e persino morte rapida. Il dietilclorofosfato (DCP) è stato riconosciuto come uno dei più noti imitatori di fattori nervosi nel combattimento chimico. Nel presente studio, presentiamo una semplice strategia per sviluppare una  striscia microporosa portatile e reversibile basata su nanocompositi per il rilevamento a occhio nudo di DCP in pochi secondi. Il cromoforo di dicianometilene diidrofuranidrazone (DCDHF-H) è stato sintetizzato mediante una facile reazione di accoppiamento azoico e incapsulato   in situ  durante la preparazione di nanocompositi di acetato di cellulosa / nanowiskers di cellulosa / idrazone (CA-CNW-H).

Questi nanocompositi CA-CNW-H hanno mostrato uno spostamento batocromico dell’intensità di assorbimento di circa 142 nm da 438 a 580 nm con l’aumento della concentrazione di DCP. L’attuale striscia sensore CA-CNW-H ha mostrato un limite di rilevamento per DCP compreso tra 25 e 200 ppm. Il cambiamento di colore di CA-CNW-H da giallo a viola dopo l’esposizione a DCP è stato rilevato dall’analisi di CIE Lab. Sono state esaminate la morfologia, la cristallinità fibrosa, la stabilità termica e le proprietà meccaniche delle strisce di sensori CA-CNW-H preparate.

Sviluppo di una test card basata su strisce immunocromatografiche d’oro colloidale per la rilevazione rapida di anticorpi contro Theileria equi e Babesia caballi.

La piroplasmosi equina (EP) è un problema serio nell’industria equina e il controllo dell’EP è fondamentale per il commercio internazionale di cavalli. L’EP è causato da due protozoi apicomplessi, Theileria equi e Babesia caballi. Metodi rapidi e accurati adatti per rilevare questi parassiti sul campo sono fondamentali per controllare l’infezione e la diffusione di EP. In questo studio abbiamo sviluppato una scheda per la rilevazione degli anticorpi contro T. equi e B. caballi basata su due strisce immunocromatografiche in oro colloidale secondo il principio del double sandwich antigen. Le proteine dell’antigene 1 (EMA1) dell’equimerozoite e la proteina rhoptry BC48 sono comunemente usate come antigeni diagnostici rispettivamente contro T. equi e B. caballi. La proteina EMA1 o BC48 purificata marcata con oro colloidale è stata utilizzata come rivelatore sulla striscia,

Il protocollo richiede da 10 a 15 minuti e non richiede apparecchiature o prodotti chimici specializzati e un test può rilevare due agenti patogeni EP su una scheda. I test di specificità hanno confermato la mancanza di reattività crociata con sieri positivi per i comuni patogeni equini. Utilizzando un kit commerciale per l’immunosorbente enzimatico (cELISA) per il confronto,  476 campioni clinici sono stati testati mediante card. I tassi di coincidenza erano rispettivamente del 96,43% e del 97,90% per T. equi e B. caballi. Il feedback delle prove sul campo è stato costantemente positivo, suggerendo che questo strumento diagnostico può essere utile per controllare la diffusione di T. equi e B. caballi.

La piroplasmosi equina (EP), causata da Theileria equi e Babesia caballi, è una grave malattia trasmessa dalle zecche nei cavalli che si verifica nella maggior parte del mondo. Il PE è considerato una malattia soggetta a denuncia dall’Organizzazione mondiale per la salute animale (OIE). La diagnosi accurata e la differenziazione di T. equi e B. caballi sono molto importanti per la prevenzione, il controllo e il trattamento dell’EP. Ecco perché abbiamo sviluppato  il test immunocromatografico Colloidal Gold Sandwich (GICG) per la rilevazione di T. equi e B. caballi. Due strisce GICG sono state impilate una accanto all’altra su una scheda di rilevamento di T. equi e B. caballi e in un test è stato possibile rilevare due agenti patogeni EP. Il metodo era semplice, veloce e specifico per la rilevazione di EP; pertanto, rispetto ai metodi precedenti, questo metodo è più adatto per la diagnostica dei patogeni sul campo.

Strisce  immunocromatografiche fluorescenti ad alta specificità nel tempo per la rilevazione simultanea di diversi pesticidi organofosforici in base alla strategia della sonda indiretta

L’uso indiscriminato di pesticidi organofosforici (PO) rappresenta un’enorme minaccia per la sicurezza alimentare, la salute umana e l’ambiente. Poiché esistono molti tipi di pesticidi, individuarli è una sfida. Il rilevamento simultaneo di più organofosfati in un test è attualmente un problema da risolvere. In questo articolo,  è stata preparata una striscia di immunocromatografia a fluorescenza a risoluzione temporale (TRFIA) utilizzando anticorpi OP ad ampia specificità (Abs) come elemento di riconoscimento. Gli addominali sono stati combinati con microsfere di lattice di ossido di europio con anticorpi anti-topo di pecora (SaMIgG) per creare una sonda indiretta.

Questa strategia può realizzare efficacemente l’amplificazione del segnale e può risparmiare la quantità e proteggere l’attività degli addominali. Al rilevamento, è stato osservato un cambiamento di colore della linea del test (linea T) per la valutazione qualitativa alla luce UV (365 nm). Le immagini campione positive sono state quindi analizzate utilizzando ImageJ per completare il rilevamento quantitativo. In condizioni di progettazione e funzionamento ottimali, il limite di rilevamento della striscia può raggiungere 0,53 ng g  ^-1  .
