    1.     Niezawodna, zautomatyzowana podręczna analiza sekwencji obrazów życiowych w mikroskopie żywym oraz identyfikacja stanów chorobowych i efektów terapii są warunkami wstępnymi rutynowego stosowania ilościowych pomiarów mikrokrążenia wgokokozy. Niniejsze badanie miało na celu kliniczną walidację niedawno wprowadzonego oprogramowania  MicroTools   w dużej wielocentralnej bazie danych pacjentów w okresie okołooperacyjnym i krytycznie chorych oraz wykorzystanie tego automatycznego algorytmu do eksploracji danych i identyfikacji zmian zmiennych mikrokrążenia podjęzykowego w odpowiedzi na chorobę i terapię.

    • L’infiammazione è accompagnata da variazioni dei livelli plasmatici dei “reagenti di fase acuta”, comprese le proteine ​​C-reattive umane e non umane. Risultati precedenti nel nostro laboratorio hanno mostrato che la CRP nel siero dei conigli in fase acuta può circolare in combinazione con lipoproteine ​​​​a densità molto bassa (VLDL), che influenzano sia il peso corporeo apparente che la mobilità elettroforetica della CRP.
    • Mostriamo ora che la capacità di VLDL di interagire con la CRP cambia nel tempo durante la risposta di fase acuta. Prima che l’infiammazione sia indotta, il fattore o i fattori responsabili della mobilità beta della PCR sono presenti a un livello basso nel VLDL. Durante le prime 24 ore della risposta di fase acuta, non è stato possibile dimostrare più attività nella frazione VLDL.

    Studio sperimentale dell’espressione di TNF-α e   CRP   nella tubercolosi spinale dopo strumentazione

    2.     Jasminum sambac  : potenziale candidato per lo sviluppo di farmaci per il trattamento delle condizioni cardiovascolari

    Il Jasminum sambac   (L.) è una pianta medicinale popolare dell’Asia meridionale tradizionalmente usata per curare i problemi cardiovascolari. La presente indagine è stata meticolosamente organizzata per indagare i fondamenti farmacologici degli usi medici di   J. sambac   nei disturbi cardiovascolari e per indagare i meccanismi sottostanti.
    • Studi meccanicistici hanno mostrato che l’estratto grezzo di foglie di   J. sambac   ha causato ex vivo un effetto vasodilatatore nella preparazione dell’anello endoteliale aortico intatto e l’effetto ipotensivo è stato registrato utilizzando trasduttori di pressione e forza collegati al sistema di acquisizione dati Power Lab.

    • L’effetto vasorilassante e cardioprotettivo in esperimenti in vivo ed ex vivo, che sono possibili attraverso l’attivazione del recettore muscarinico e/o il rilascio di ossido nitrico e la riduzione dell’adrenalina, stress ossidativo indotto, ne giustifica l’uso nei disturbi cardiovascolari.

    Presenza di infezione da tularemia in pazienti con febbre maculata giapponese

    1. Nel nostro studio precedente, IFN-ɤ e IL-6 hanno dimostrato di essere citochine immunitarie chiave contro l’infezione da Rickettsia japonica (R. japonica) misurando i livelli sierici di citochine nella fase acuta e curabile della febbre maculata giapponese (JSF). La tularemia è una malattia infettiva causata da una puntura di zecca o dal contatto con animali infetti, nota anche come  febbre di coniglio. Nessun caso confermato è stato identificato in Giappone negli ultimi due decenni.
    2. Espressione genica condizionale mediata da lentivirus
La capacità di identificare il ruolo di un particolare gene in   un sistema   dipende dal controllo dell’espressione di quel gene. In questo protocollo descriviamo un metodo per l’espressione condizionale stabile dei recettori Nod-Like (NLR) nelle cellule THP-1 utilizzando un   sistema di   espressione  lentivirale  . Questo sistema  combina tutti i componenti necessari per l’espressione genica indotta dalla tetraciclina in un singolo lentivettore con la coespressione costitutiva di un marcatore selezionabile, che è un modo efficiente per controllare l’espressione genica utilizzando una singola infezione virale delle cellule. Questo viene fatto sulla piattaforma di espressione  di lentivirus di terza generazione  , che migliora la sicurezza dei lentivirus e consente una maggiore espressione genica rispetto alle piattaforme precedenti lentivirale.
  • Il plasmide di espressione  del lentivirus   viene prima progettato per contenere il gene di interesse guidato dal promotore TRE (elemento di risposta alla tetraciclina) in una semplice fase di clonazione del gate, e quindi co-trasfettato nelle cellule HEK293T insieme ai   plasmidi di  confezionamento e involucro per   produrre il virus.
  • Il virus viene utilizzato per infettare il tipo cellulare di interesse a un MOI basso tale che la maggior parte delle cellule trasdotte contenga una singola integrazione virale. Le cellule infette vengono coltivate con la selezione e l’integrazione del virus viene verificata mediante qPCR.
  • L’espressione genica nelle cellule trasdotte stabilmente è indotta dalla doxiciclina e convalidata da qPCR, immunoblotting e citometria a flusso.

Impatto minimo di ZAP sulla  produzione di vettori lentivirali   e sull’efficienza di trasduzione

  1. La proteina ZAP antivirale lega i dinucleotidi CpG sull’RNA virale, inibendo la replicazione. Ciò probabilmente ha portato alla soppressione di CpG osservata in molti virus a RNA, inclusi i retrovirus. Le sequenze aggiunte ai genomi dei vettori retrovirali, come promotori interni, transgeni o elementi regolatori, aumentano notevolmente l’abbondanza di CpG. Poiché questi CpG possono consentire a ZAP di indirizzare il vettore RNA retrovirale, abbiamo analizzato se questa riduzione della produzione del vettore, dell’efficienza di trasduzione e dell’espressione del transgene.
  2. Sorprendentemente, sebbene l’HIV-1 ad alto contenuto di CpG sia stato effettivamente inibito da ZAP nelle cellule HEK293T, l’esaurimento di ZAP non ha aumentato significativamente il titolo del   vettore lentivirale utilizzando diversi    plasmidi genomici e  di confezionamento . Anche la sovraespressione di ZAP non ha inibito il titolo del  vettore lentivirale. Inoltre, una riduzione dell’abbondanza di CpG nel genoma del  vettore lentivirale   non ha aumentato il suo titolo  e il vettore gammaretrovirale derivato dal virus della leucemia murina non è stato significativamente limitato da ZAP.
  3. Nel complesso, dimostriamo che la maggiore quantità di CpG nei vettori retrovirali rispetto ai retrovirus di tipo selvaggio da cui derivano non li rende intrinsecamente sensibili allo ZAP. Un’ulteriore comprensione del modo in cui ZAP mira specificamente alle trascrizioni per inibire la loro espressione può consentire lo sviluppo di contesti di sequenza CpG che reclutano o aggirano con successo questo   sistema  antivirale .

Guida alla  produzione di vettori lentivirali   per cellule difficili da trasfettare utilizzando cellule che esprimono c-kit derivate dal cuore come   sistema modello

  • La terapia genica è la modifica del corredo genetico mediante l’introduzione di acidi nucleici estranei nelle cellule bersaglio attraverso metodi di rilascio genico per il trattamento di una specifica malattia.
  • Sebbene i geni bersaglio svolgano un ruolo chiave nella terapia genica, anche il  sistema di rilascio del gene utilizzato  è di grande importanza nel determinare il successo della terapia genica. Poiché le cellule primarie e le cellule staminali sono spesso bersagli della terapia genica negli studi clinici,   il sistema di somministrazione  dovrebbe essere robusto e le voci basate su virus come i  vettori lentivirali   funzionano meglio per trasportare il transgene nelle cellule. Tuttavia, anche all’interno  dei vettori lentivirali   , diversi parametri possono influenzare la funzionalità   del sistema di rilascio  .
  • Utilizzando cellule che esprimono c-kit (CC) derivate dal cuore come sistema modello   ,  il presente studio mira a ottimizzare la  produzione lentivirale   studiando vari fattori sperimentali come la  produzione del sistema lentivirale  , il  metodo di concentrazione e il tipo di marcatore selezionabile. I nostri risultati hanno mostrato che il sistema di seconda generazione   con   pCMV-dR8.2 dvpr come  plasmide di confezionamento ha comportato   un’efficienza di produzione  di lentivirus 7,3 volte superiore  rispetto a psPAX2.
  • La concentrazione del virus dell’ultracentrifuga ha determinato un titolo del virus più elevato a oltre 5×105  unità infettive   /ml (IFU/ml). Infine, la concentrazione inibitoria minima (MIC) del marcatore di selezione della puromicina era di 10 µg/ml e 7 µg/ml per HEK293T e CC, indicando l’utilità della selezione di antibiotici per tutti i tipi cellulari. Questi dati incoraggianti possono essere estrapolati e applicati ad altre cellule difficili da trasfettare, come diversi tipi di cellule staminali o cellule primarie.

  1. L’efficacia del nuovo metodo PCR con una sonda di spegnimento nel rilevare le mutazioni dell’rRNA 23S nei bambini con infezione da Mycoplasma pneumoniae
    • Recentemente è stato sviluppato un kit diagnostico molecolare rapido automatizzato (Smart Gene Myco) per la rilevazione individuale dei geni di Mycoplasma pneumoniae (MP). Questo nuovo approccio ai test non richiede attrezzature o abilità speciali e può essere completato in 50 minuti. Abbiamo valutato prospetticamente questo kit diagnostico, insieme ad altri test convenzionali, per la diagnosi di polmonite nei bambini. Campioni di 98 bambini  (50 ragazzi e 48 ragazze; età 1-14; media: 4,7 ± 2,1 anni; mediana: 4 anni) con diagnosi clinica di polmonite sono stati testati per MP utilizzando la reazione a catena della polimerasi nel tempo reale (RT-PCR) come metodo di riferimento  . I risultati di tre test diagnostici molecolari, anticorpi anti-MP sierici e colture MP sono stati confrontati con i dati RT-PCR.
    • Tra 98 bambini, 38 sono risultati positivi al MP. Tutti i risultati della diagnostica molecolare hanno mostrato pieno accordo con i dati RT-PCR. La sensibilità della coltura era del 64%, mentre la sensibilità dei kit ImmunoCard Mycoplasma e SERODIA Myco II era inferiore (39% e 29% rispettivamente). Inoltre, è stata trovata una correlazione positiva significativa tra il numero di copie MP e la sensibilità del test colturale (r = 0,95, p = 0,048). Mutazioni di resistenza ai macrolidi nel gene dell’RNA ribosomiale 23S sono state rilevate in 24 su 38 bambini con lo Smart Gene Myco basato sulla PCR con la sonda di spegnimento, confermate dal sequenziamento diretto, rivelando tutte le mutazioni come A2063G. Questo è il primo studio per valutare l’utilità clinica dello Smart Gene Myco Kit.

    Rilevazione rapida e specifica delle infezioni da Mycoplasma gallisepticum e Mycoplasma synoviae nel pollame mediante test PCR singoli e duplex.

    La micoplasmosi aviaria, causata principalmente da Mycoplasma gallisepticum (MG) e Mycoplasma synoviae (MS), è una malattia economicamente importante dell’industria avicola. Lo scopo di questo studio era sviluppare la PCR duplex come metodo rapido, specifico ed economico per rilevare accuratamente MG e MS nel pollame e confrontarlo con una singola PCR MG/MS (monoplex). Nel presente studio sono stati testati un totale di 146 allevamenti di pollame con una storia clinica di malattie respiratorie. Campioni di tessuto in pool (trachea, polmone e sacche d’aria) da 3 a 4 uccelli da ciascun gregge sono stati raccolti durante l’autopsia presso i Disease Research Laboratories, Hisar, Haryana, India. I test PCR singoli e duplex sono stati standardizzati utilizzando primer intergeniciregione spaziatrice (IGSR; 16S-23S rRNA) per il gene MG ed emoagglutinina vlhA per la SM, con una dimensione prevista dell’amplicone rispettivamente di 812 bp e 1200 bp  .
    In una singola reazione PCR, campioni di tessuto del 6,85%, 2,74% e 2,74% erano positivi rispettivamente per MG, MS e sia MG che MS. Tuttavia, la PCR duplex ha mostrato rispettivamente il 7,53%, il 2,74% e l’1,37% di positività per MG, MS e sia MG che MS. Prendendo i risultati della PCR monoplex come gold standard, la sensibilità e la specificità della PCR duplex sviluppata erano rispettivamente del 94,44% e del 100%. Inoltre, la statistica kappa di Cohen (k = 0,97) ha misurato un accordo “eccellente” tra i test PCR monoplex e duplex. I valori predittivi positivi e negativi della PCR duplex sono risultati rispettivamente di 1,0 e 0,9922, con un intervallo di confidenza (CI) del 95%, rispetto alla PCR monoplex. L’uso simultaneo di due geni in una reazione PCR duplex era più veloce ed economico di due reazioni PCR singole separate.

Isolato da  campioni clinici sottoposti a un laboratorio diagnostico. Dei 1.548 campioni dall’infezione della mammella, latte dalle vasche di raccolta e fluidi biologici, sono stati ottenuti 38 isolati di micoplasma. M. bovis, M. canadense, M. californicum e M.leachii sono stati rilevati da due reazioni multiplex PCR, confermando la loro presenza clinica nei bovini da latte. Le tecniche utilizzate in questo studio potrebbero essere utili per migliorare la nostra comprensione delle infezioni da Mycoplasma nei bovini, poiché la ricerca di questi organismi di solito non è inclusa nelle diagnosi di routine.

    Utilità della   PCR Q-probe   nei bambini con  infezione da Mycoplasma   pneumniae

    Per studiare l’utilità della reazione a catena della polimerasi di spegnimento della sonda (Q-probe PCR) per il rilevamento di Mycoplasma pneumoniae (MP) resistente ai macrolidi, abbiamo analizzato retrospettivamente il decorso clinico di 21 bambini con infezione da MP. La percentuale di MP resistenti ai macrolidi era del 66,7%. La durata della febbre dopo il trattamento antibiotico iniziale era più lunga nei pazienti con infezione da MP resistente ai macrolidi rispetto ai pazienti con infezione da MP sensibile ai macrolidi. La durata della febbre dopo la PCR Q-probe non differiva significativamente tra i pazienti con infezione da MP resistente ai macrolidi e pazienti con infezione da MP sensibile ai macrolidi. Questi risultati suggeriscono che l’uso di antibiotici basati sulla PCR Q-probe ridurrà la durata della febbre. Q-probe PCR contribuisce alla determinazione di antibiotici appropriati e al miglioramento del decorso clinico dell’infezione da MP.

    Sviluppo di un nuovo  test PCR   con SYBR Green I per la rilevazione di   Micoplasma  , Acoleplasma  e   Ureaplasma sp.in   colture cellulari

    1. Mycoplasma  ,   Acholeplasma  e   Ureaplasma   sp. Sono batteri atipici responsabili della contaminazione delle colture cellulari in vitro, che possono falsare i risultati. Questi batteri causano anche infezioni negli esseri umani e negli animali e possono portare a malattie croniche. Nella reazione a catena della polimerasi (PCR) sviluppata in questo studio, è stata utilizzata la PCR quantitativa con il fluorocromo SYBR Green I per facilitare il   rilevamento e l’identificazione del DNA di Mycoplasma  ,   Acholeplasma  e   Ureaplasma   sp.
    2. Il test di screening-1 v.1 (triplo qPCR) ha rilevato 11 specie. Test-1 v.2 (tre singole qPCR) inizialmente ha identificato tre sottogruppi, limitando così l’uso di singole qPCR  nel Test-2 per l’identificazione delle specie. Lo scopo di entrambi i test era in linea con i requisiti della farmacopea per il controllo della qualità microbica delle cellule di mammifero e includeva il rilevamento di   M.  arginini, M. orale, M. hyorhinis,   M.  fermentans   ,  M.   genitalium  ,   M.  hominis   ,  M.   pneumoniae  ,   m. salivarium  ,   M. pirum  ,   A.  laylawii e   U. urealyticum. Il limite di rilevamento variava da 125-300 a 50-100 copie per millilitro rispettivamente nel test-1 e nel test-2.
    3. Il test-1 e il test-2 hanno mostrato risultati completamente coerenti, risparmiando tempo per il rilevamento e/o l’identificazione di specie selezionate di   micoplasma  ,   acoleplasma  e   ureaplasma   nelle colture cellulari testate.

Predizione   di Mycoplasma   pneumoniae e Chlamydophila pneumoniae nella polmonite acquisita in comunità (CAP): uno studio epidemiologico delle infezioni respiratorie  mediante saggi PCR multiplex

  • La polmonite acquisita in comunità (PAC) è una malattia comune che può portare alla mortalità. I β-lattamici sono inefficaci contro i patogeni atipici, compreso il Mycoplasma pneumoniae. Abbiamo utilizzato studi molecolari per sviluppare un albero decisionale per predire i patogeni CAP atipici e per studiare la prevalenza della resistenza ai macrolidi in Mycoplasma pneumoniae.
  • Abbiamo condotto uno studio osservazionale prospettico su pazienti di età ≥ 18 anni con febbre e sintomi respiratori a cui è stata diagnosticata la CAP in uno dei due ospedali comunitari tra dicembre 2016 e ottobre 2018.
  • Abbiamo valutato  le combinazioni di variabili cliniche che meglio predicevano i patogeni atipici associati alla CAP mediante l’analisi dell’albero di classificazione e regressione (CART) . La polmonite è stata definita come sintomi respiratori e nuova infiltrazione diagnosticata mediante radiografia del torace o TC del torace.
  • Abbiamo analizzato 47 pazienti (21 donne, 44,7%, età media 47,6 anni). Patogeni atipici sono stati rilevati in 15 pazienti (31,9%; 12 Mycoplasma pneumoniae, 3 Chlamydophila pneumoniae). Dieci pazienti avevano Mycoplasma pneumoniae resistente ai macrolidi (tasso di resistenza ai macrolidi 83,3%). L’analisi CART ha suggerito che i fattori associati alla presenza di agenti patogeni atipici erano non cracking, età <45 anni e LD ≥ 183 U/L (sensibilità 86,7% [59,5, 98,3], specificità 96,9% [83,8, 99,9]).
  • I nostri semplici principi decisionali clinici possono essere utilizzati per identificare i pazienti delle cure primarie con CAP che sono a rischio di patogeni atipici. Sono necessarie ulteriori ricerche per confermare la sua idoneità a diverse popolazioni.Registrazione dello studio Studio clinico (ID studio UMIN: UMIN000035346).

Confronto di tre  metodi PCR in tempo reale per il rilevamento del Mycoplasma   genitalium  resistente ai macrolidi   in Svezia

L’aumento della resistenza ai macrolidi dei ceppi di Mycoplasma genitalium è osservato in tutto il mondo, il che ha un grave impatto sul trattamento. Lo scopo di questo studio era confrontare tre metodi PCR in tempo reale per rilevare la resistenza ai macrolidi: il metodo PCR proprietario descritto da Touati et al., ResistancePlus MG (SpeeDx) e S-DiaMGRes ™ (Diagenode Diagnostics). Sono stati analizzati cento  campioni di pazienti svedesi positivi per M. genitalium e  il controllo quantitativo del DNA di M. genitalium.
La resistenza ai macrolidi è stata rilevata in 18, 15 e 16 campioni con metodi appropriati. Il sequenziamento del gene 23S rRNA ha confermato la resistenza in 16 (16%) dei 100 campioni in cui la resistenza è stata rilevata con uno qualsiasi dei tre metodi.

Test dell’attività proteasica nell’intestino di una mosca
      • L’intestino è l’organo centrale necessario per digerire il cibo, assorbire i nutrienti e combattere gli aggressori ingeriti con il cibo. L’alterazione della fisiologia intestinale a seguito di danni alla mucosa   influisce sulla sua capacità digestiva, che a sua volta influenzerà la crescita, il benessere e persino la sopravvivenza dell’individuo. 
      • Pertanto, la valutazione della funzione intestinale comprende, tra l’altro, il monitoraggio della sua integrità, il rinnovamento cellulare, la sua difesa immunitaria, la produzione di ormoni enteroendocrini e la sua capacità digestiva. Qui descriviamo in dettaglio come valutare l’attività delle proteasi secrete nel lume della   mosca adulta Drosophila melanogaster  . Questo metodo può essere applicato anche agli intestini larvali. Il presente protocollo è adattato e migliorato dal protocollo Sigma-Aldrich proposto nel “Protease Fluorescence Detection Kit” (codice prodotto PF0100).

      Test in vitro  di    scissione dell’istone H3    per proteasi  H3 di lievito e fegato di pollame 

      1. Le proteine ​​istoniche subiscono un’ampia varietà di modificazioni post-traduzionali (PTM) reversibili e irreversibili (Bannister e Kouzarides, 2011; Azad e Tomar, 2014). È noto che i PTM sugli   istoni regolano la struttura e la funzione della cromatina. Gli istoni sono irreversibilmente modificati dal troncamento proteolitico dei loro domini di coda   .
      2. Il troncamento proteolitico delle code dell’istone continua ad attirare l’attenzione dei ricercatori nella biologia della cromatina. Possiamo riassumere il troncamento di H3 eseguendo un test di   scissione di H3 in vitro. Qui forniamo un protocollo dettagliato per l’esecuzione di un   test di scissione H3 in vitro.

      Le analisi spettrometriche di massa rivelano discrepanze nei profili di inibizione per la proteasi simile a SARS-CoV-2.

      1. Le due proteasi SARS-CoV-2, ovvero la proteasi maggiore (M pro) e la proteasi simil-papina (PL pro), che idrolizzano la catena polipeptidica virale per dare proteine ​​funzionali non strutturali, sono essenziali per la replicazione virale e sono mediche obiettivi di chimica. Vi presentiamo un test di spettrometria di massa (MS) ad alto rendimento che   monitora direttamente la catalisi pro PL in vitro. Il test è stato utilizzato per studiare l’effetto degli inibitori PL pro di piccole molecole segnalati e degli inibitori M pro selezionati sulla catalisi PL pro.
      2. I risultati mostrano che alcune, ma non tutte, le potenze degli inibitori di PL pro differiscono significativamente da quelle ottenute utilizzando saggi basati sulla fluorescenza. Alcuni inibitori M pro concorrenti del substrato, in particolare PF-07321332 (nirmatrelvir), che è in fase di sperimentazione clinica, non inibiscono PL pro. Inibitori meno selettivi di M pro, ad esempio auranofin, inibiscono PL pro, evidenziando la possibilità di una doppia inibizione di PL pro / M pro. I test PL pro basati su MS, che sono ortogonali ai test basati sulla fluorescenza comunemente usati, sono utili per convalidare la potenza degli inibitori, in particolare per gli inibitori  basati su meccanismi non covalenti.

      Saggio di   inclusione   dell’inibitore della proteasi   dell’HIV-1 

      • Il virus minaccia continuamente la nostra vita e la nostra salute. Tra questi, l’HIV ha causato milioni di morti dalla sua scoperta.
      • La ricerca sulla selezione degli inibitori virali è diventata un’esigenza urgente. È stato sviluppato un test affidabile per l’inclusione nella selezione di un inibitore della proteasi dell’HIV-1 (HIV-1 PR).
      • Il test di fattibilità del rilevamento della proteasi è stato dimostrato sulla base del trasferimento di energia di risonanza di fluorescenza tra ossido di grafene e peptidi modificati con colorante.
      • Oltre al rilevamento rapido e sensibile dell’HIV-1 PR, sono stati misurati anche i parametri cinetici dell’enzima. Soprattutto, questo test è stato utilizzato per distinguere tra l’efficacia dell’inibitore della proteasi e la selezione di un inibitore sconosciuto.

      Nuovo    test    per la determinazione della  proteasi di precisione mediante elettroforesi capillare

      1. Il rilevamento dell’attività della proteasi nel corpo gioca un ruolo importante nella diagnosi precoce delle malattie. Tuttavia, gli enzimi entrano inevitabilmente in contatto con i vari fluidi biologici complessi nel corpo durante il flusso, il che aumenta notevolmente la difficoltà di rilevamento. Pertanto, il rilevamento della proteasi in vivo è associato a grandi sfide. In questo documento, descriviamo   un nuovo test di rilevamento della proteasi utilizzando l’elettroforesi capillare all’interno di un capillare con curve semicircolari  . In primo luogo, abbiamo progettato un substrato peptidico, quindi il peptide è stato autoassemblato con punti quantici per formare il substrato peptidico QDs.
      2. Il capillare è stato piegato in bobine semicircolari e fungeva da microreattore consentendo alla proteasi e al substrato di reagire al suo interno. A causa della diversa velocità elettroforetica, la proteasi e il substrato sono stati miscelati all’interno del capillare piegato con successive iniezioni e il taglio del substrato potrebbe essere rilevato mediante elettroforesi capillare combinata con la tecnologia di trasferimento dell’energia di risonanza di Förster. Questo nuovo test estenderà significativamente il rilevamento dell’attività enzimatica in vivo.

      Convalida e invalidazione dei principali   inibitori della proteasi SARS-CoV-2 utilizzando  i saggi della luciferasi  Flip-GFP e    Protease   -Glo 

      • La principale proteasi di SARS-CoV-2 (M   pro   ) è uno dei bersagli farmacologici più utilizzati per COVID-19. Composti strutturalmente diversi sono stati descritti come inibitori di Mpro, il che solleva la questione della loro specificità target   . 
      • Per chiarire la specificità del bersaglio e il coinvolgimento del bersaglio cellulare degli inibitori dichiarati, abbiamo  caratterizzato sistematicamente il loro meccanismo d’azione mediante il test FRET senza cellule, il test di legame dello spostamento termico, il test del lisato cellulare della luciferasi Protease-Glo e il test saggio FlipGFP cellulare.
      •  Collettivamente, i nostri risultati hanno mostrato che la maggior parte degli inibitori di M   pro   identificati durante il riutilizzo di farmaci, inclusi ebselen, carmofur, disulfiram e shikonin, sono inibitori della cisteina promiscua che non sono specifici di M   pro   , mentre clorochina, ossitetraciclina, montelukast, candesartan, e il dipiridamolo non ha inibito M   pro    in nessuno dei test testati. Nel complesso, il nostro studio evidenzia la necessità di una rigorosa convalida dei risultati all’inizio della scoperta di farmaci.

      Test in silico    e    in    vitro    rivelano potenziali inibitori della 3CL    pro   –    proteasi principale    SARS-CoV-2

      1. La pandemia di COVID-19, causata dal nuovo coronavirus della sindrome respiratoria acuta 2 (SARS-CoV-2), non mostra segni di rallentamento, anche dopo i continui sforzi per la vaccinazione. I pericoli dei nuovi ceppi sono preoccupanti poiché alcuni di essi sono più contagiosi dei ceppi originali. Pertanto, è urgentemente necessario un trattamento contro la malattia. Qui stiamo usando il database DrugBank per selezionare potenziali inibitori contro la proteasi 3CL   pro    SARS-CoV-2 principale. Invece di utilizzare il   tradizionale approccio di screening di docking computazionale, abbiamo sviluppato un regressore di cresta nucleare (utilizzando alcuni dati di docking) per prevedere le energie di legame del ligando.
      2. Abbiamo utilizzato questo modello per cercare nel database DrugBank e un elenco di due candidati principali (bromocriptina e avoralstat) per il   test enzimatico in vitro. I nostri risultati mostrano che l’attività enzimatica di 3CL   pro    in presenza di 100 μM di bromocriptina e concentrazioni di avoralstat è rispettivamente del 9,9% e del 15,9%   . Sorprendentemente, la bromocriptina ha mostrato un IC50 submicromolare di   130    nM (0,13    µM   ). Avoralstat ha mostrato un IC50   di    2,16 µM. Inoltre, le interazioni di entrambi i farmaci con 3CL   pro   sono state   analizzate utilizzando simulazioni di dinamica molecolare a 100 ns. I risultati indicano che entrambi i ligandi sono  stabili nella tasca di rilegatura 3CL   pinza  professionale .
